Introducing a sample solution that includes a target nucleic acid polymer into a microfluidic device that has a sample region with: (i) a flow channel in fluid communication with one fluid inlet and one fluid outlet; and (ii) a plurality of valves that when closed, partition the flow channel into isolated reaction sites. Each valve is formed from an elastomeric membrane that separates a control channel from the flow channel and which is disposed to be deflected into or retracted out from the flow channel in response to an actuation force to the control channel thereby forming the valve.
