We describe a strategy for designing protein-fragment complementation assays to detect protein-protein interactions in vivo. The design and implementation of this strategy is illustrated with the enzyme murine dihydrofolate reductase (DHFR). Fusion peptides consisting of N- and C-terminal fragments of murine DHFR fused to GCN4 leucine zipper sequences were coexpressed in Escherichia coli grown in minimal medium where the endogenous DHFR activity was inhibited with trimethoprim. Coexpression of the complementary fusion products restored colony formation. Survival only occurred when both DHFR fragments were present and contained leucine-zipper forming sequences demonstrating that, …

Why it's free to use
- Filed January 31, 1997. No grant is recorded for it in Canada; had it been granted, its 20-year term would have ended January 31, 2017.
- A certificate of supplementary protection adds at most two years for a drug, so even that would have ended by January 31, 2019.
- We check Canadian rights only. A patent in its family in another country could still be in force: check its family on Google Patents.
