A method of quantitatively estimating, biologically-active cyclosporines analogues, derivatives and metabolites by a competitive protein binding assay using water-soluble binding proteins isolated from the cycloplasm of human and animal cells. Tracers include radioactive atoms fluorescent, molecules, enzymes, and chemiluminescent molecules. Solution and solid phase assay methods are disclosed. Also claimed are the water-soluble binding proteins.
